It is possible to transfer a large number of families to a newer version or it has to be done family by family?

As long as I know, It has to be one by one. You can´t update them all at the same time. But maybe You could update all the families within a project when the project itself is updated. But the next step would be to save all the families in other location for not get confused with the old ones.

Yes, it's possible to transfer a large number of families to a newer version of Revit. You can use batch conversion tools or scripts to automate the process, saving time and effort compared to transferring each family individually.
