I am new to Revit Material Take Offs and i am struggling to figure out how to calculate the qty of linear feet of interior door casing.

I would ultimately like to calculate the qty by molding profile as we often use multiple profiles in a residential project in an effort to match existing profiles in an existing home

Why????

I would use the door size and add for waste and joint.  2H + W + waste.

Can I do that in the calculations field so it does it automatically?

How do I differentiate between multiple profiles being used?

You canj add a calculated field to your schedule.  If you have different profile types, you can add that as a type parameter to doors and then sort by that in your schedule
