Hi, I am new to Revit so this is probably a basic question. 

I am building a church with some unique walls.

 

Is the best way to first build a generic wall then edit those walls? (Obviously i haven´t learn the restrictions/flexibility in generic walls)

Or should i make an In-place family for the walls?

You have not told us enough about the walls to answer your question.  A wall profile sketch would help.
